Nonsense-mediated mRNA decay, a simplified view for a complex mechanism

The nonsense-mediated mRNA decay (NMD) mechanism is both a quality control mechanism and a gene regulation pathway. NMD has been studied for more than 30 years with an accumulation of many mechanistic details which have often led to debate leading to the proposal of several models of NMD activation, in particular in higher eukaryotes. Two models in particular seem to be opposed since the first requires the intervention of the protein complex EJC (exon junction complex) to recruit NMD factors downstream of the premature termination codon (PTC), when the second model is independent of the 'EJC and indicates that NMD factors concentrate in the 3'UTR part to initiate the NMD reaction in the presence of a PTC. We describe in this review both models giving recent molecular details and providing experimental arguments supporting one or the other model. In the end it is certainly possible to imagine that these two models co-exist rather than thinking that they are exclusive.
